Step 2; To a solution of 2-methanesulfonyl-pyrimidine-5-carboxylic acid methyl ester 2 (2.5 g, 11.56 mmol) in DCM (30 mL) is added a solution of tetrabutylammonium cyanide (3.1 g, 11.56 mmol) in water (30 mL) slowly at room temperature. The mixture is stirred for 80 min. The mixture is washed with water (2x20mL), dried (Na2SO4), filetered and concentrated in vacuo. The residue is purified by column chromatography eluting with 5-60% EtOAc in heptane to afford 2-cyano-pyrimidine-5- carboxylic acid methyl ester (1.16 g, 61%) as a solid. MS: 164 (M+H); 1HNMR (300 MHz, CDCl3): δ 4.05 (s, 3H), 9.37 (s, 2H).
