4.2 4. Synthesis of isocyanides

General procedure: The amine (1 mmol) is dissolved in ethyl formate, in the presence of base (Et3N or DIPEA) when necessary, and the solution is refluxed overnight at 70 - 80 oC. The solvent is removed under reduced pressure and the corresponding formamide (checked by TLC) is purified when necessary and dissolved in dry DCM or THF. Et3N (5 mmol) and POCl3 (1 mmol) - drop wise and over 15 min - are then added under nitrogen atmosphere at 0 °C and the reaction mixture is allowed to reach room temperature and stirred for 2 to 3 additional hours until completeness. A saturated solution of NaHCO3 is added drop wise to neutralize and quench the reaction and the organic phase is separated and washed twice with brine. The volatiles are removed under reduced pressure and the product is immediately purified by column chromatography and stored under nitrogen atmosphere at -20 oC.
