Niclosamide, 2,5-dichloro-4nitrosaicylanilide (38.1.34), is made by reacting 5-chlorosalicylic acid with 2-chloro-4-nitroaniline in the presence of phosphorus trichloride.

Steps:

1 Synthesis step 1: preparation N-(2-chloro-4-nitro-phenyl) - 5-chloro-2-hydroxy-benzamide

In 135 °C, to the 5- chlorine salicylic acid (3.44g, 20 . 0mmol) and 2-chloro-4-nitroaniline (3.44g, 20 . 0mmol) dissolved in chlorobenzene mixing solution of adding dropwise PCl3(2.4g, 17 . 5mmol) dissolved in a chlorobenzene solution. 3 hours later, the reaction solution is cooled to the room temperature. Filtering and collecting the resulting solid, wash, then from ethyl acetate (or acetone) recrystallization to obtain a white solid (6.8g, 68.7%) shaped N-(2-chloro-4-nitro-phenyl) - 5-chloro-2-hydroxy-benzamide.
