PIPERIDIN-4-YL-(3-TRIFLUOROMETHYL-PHENYL)-AMINE synthesis

Reaction Conditions:

Stage #1: tert-butyl 4-((3-(trifluoromethyl)phenyl)amino)piperidine-1-carboxylatewith trifluoroacetic acid in dichloromethane at 20; for 2 h;
Stage #2: with sodium hydroxide in water; pH=14;

Steps:

Description 21: N-[3-(Trifluoromethyl)phenyl]-4-piperidinamine (D21 ). A solution of two combined preparations of D20 (0.86g, 2.5mmol) in DCM (40ml) was treated with TFA (10ml) and the reaction stirred at room temperature, under argon for 2h. The solvent was removed in vacuo and the residue partitioned between DCM and water. The aqueous was basified to pH14 with 2M NaOH solution then extracted with EtOAc (x5). The combined organics were dried (Na₂SO₄) and concentrated in vacuo to give the title compound as a transparent oil (0.64g). δ_H (CDCI₃, 250MHz) 7.24 (1H₁ t), 6.90 (1H₁ d), 6.74 (2H₁ m), 3.75 (1H₁ m), 3.40 (1H₁ m), 3.14 (2H, m).2.74 (2H₁ m), 2.07 (2H, m), 1.34 (2H₁ m).
