Reaction Conditions:

Stage #1:5,15,10,20-tetraphenylporphyrin with sulfuric acid at 100; for 8 h;
Stage #2: with sodium hydroxide in water; pH=9

Steps:


H2TPP (1 g) and 25 mL of H2SO4 (98%) were stirred for 8 h at 100 oC. The solution was then left overnight at room temperature, filtered, and washed with a small quantity of water, after which 150 mL of hot water was added and neutralized by slow addition of 0.1 M NaOH to pH of 9in ice bath. The sodium sulfate salt was filtered and washed many times with methanol to extract the compound. The solution was evaporated to dryness and purified by reverse phase column chromatography using water: methanol (95:5) solvent system to afford highly water soluble purple crystal, with 91% yield. 1H NMR (400 MHz, DMSO-d6) δ: 8.85 (s, 8H, H-pyrrolic), 8.18 (d, 8H, m-phenyl), 8.06 (d, 8H, o-phenyl), -2.93 (s, 2H, NH) ppm; UV-vis (λmax in H2O): 414, 520, 552, 581, 635 nm.
