히스티딘에이치씨엘 C화학적 특성, 용도, 생산

순도시험

  (1) 용상 : 이 품목 1g을 물 10mL에 녹일 때, 무색으로서 그 탁도는 거의 징명 이하이어야 한다.

  (2) 액성 : 이 품목의 수용액(1→10)의 pH는 3.5～4.5이어야 한다.

  (3) 비선광도 : 이 품목을 98～100℃에서 3시간 건조한 다음 약 5.5g을 정밀히 달아 6N 염산에 녹여 50mL로 하여 이 액의 선광도를 측정할 때, ＝+8.5～+10.5°이어야 한다.

  (4) 비소 : 이 품목을 비소시험법에 따라 시험할 때, 그 양은 4.0ppm 이하이어야 한다.

  (5) 납 : 이 품목 5.0g을 취하여 원자흡광광도법 또는 유도결합플라즈마발광광도법에 따라 시험할 때, 그 양은 5.0ppm 이하이어야 한다.

확인시험

  (1) 이 품목의 수용액(1→10)에 수산화나트륨용액(1→4)을 가하여 알칼리성으로 하면 좌선성이나 이를 염산으로 산성으로 하면 우선성으로 변한다.

  (2) 이 품목의 수용액(1→1,000) 5mL에 닌히드린시액 1mL를 가하여 3분간 가열하면 자색을 나타낸다.

  (3) 이 품목은 확인시험법 중 염화물 (나)의 반응을 나타낸다.

  (4) 이 품목의 수용액(1→100) 5mL에 브롬시액 2mL를 가하면 액은 황색을 나타내고 조용히 가열하면 무색으로 되었다가 적갈색을 거쳐 흑색 비슷한 색의 침전이 생긴다.

정량법

  이 품목을 건조한 다음 약 0.2g을 취하여 이하 「L-라이신염산염」의 정량법에 따라 정량한다.

0.1N 과염소산용액 1mL ＝ 10.48mg C₆H₉O₂N₃•HCl•H₂O

강열잔류물

  이 품목의 강열잔류물은 0.1% 이하이어야 한다.

용도

L-Histidine is used in cell culture media formulations used in biomanufacturing. L-Histidine has been used to study cultures of the human T-lymphoblastic leukemia cell line MOLT-4 to study modulation of apoptosis.

Purification Methods

Crystallise the monohydrochloride from aqueous EtOH or 60% aqueous EtOH (m 259odec). Alternatively dissolve 10g in 50mL of H2O, decolourise with Norite, filter, evaporate it in a vacuum to a syrup, cool to room temperature, add 95% EtOH with stirring until slightly turbid, scratch the sides of the vessel until crystals form, then add slowly 40mL of EtOH and keep at 0o overnight, filter the solid off, wash it several times with EtOH and dry it in a vacuum.
