Reaction Conditions:

with potassium permanganate in acetone at 0 - 20; for 3 h;

Steps:

4.2 5-Bromothiophene-2-carboxylic acid (7)
To a solution of compound 6 (2 mmol) in acetone (15 mL) was added KMnO4 (0.38 g, 2.4 mmol) in batches at 0 °C. The mixture was stirred at r.t. for 3 h. After it was fully reacted, the mixture was filtrated and filtrate was concentrated under vacuum. The residue was purified by column chromatography. Yield: 85%; 1H NMR (500 MHz, CDCl3) δ 7.64 (d, J = 4.5 Hz, 1H), 7.11 (d, J = 4.5 Hz, 1H). ESI-MS: m/z = 205 [M - H]-.
