4-(氯甲基)-2-甲基-1,3-噻唑性质、用途与生产工艺

简介

4-(氯甲基)-2-甲基-1,3-噻唑是一种烷基化的噻唑类衍生物，在高温下容易发生分解反应，可用作有机合成基础原料常用于在目标有机分子结构中引入噻唑单元，广泛地应用于噻唑类HIV 蛋白酶抑制剂的合成。

制备

在一个干燥的1升反应器中将 1,3-二氯丙-2-酮（12.7 克）和硫代乙酰胺（7.5 克）在干燥乙醇（70 毫升）中的溶液在氩气环境下加热至回流搅拌反应 5.5 小时。反应结束后将反应混合物冷却至室温，然后将其在减压下进行浓缩，得到的深色结晶残留物溶于水（250 mL）中，然后往上述水溶液中缓慢的加入固体 NaHCO3 使反应粗体系的pH 值≈8。然后用Et2O（4 x 100 mL）萃取混合物，并将合并的有机溶液在无水MgSO4 上进行干燥处理。过滤除去干燥剂并将所得的滤液在真空下进行浓缩，最后通过减压蒸馏（压力为0.05 托）提纯残留物即可得到目标产物分子4-(氯甲基)-2-甲基-1,3-噻唑。
