草酸萘呋胺(Naftidrofuryl Oxalate):C24H33NO3?C2H2O4。[3200-06-4]。从乙酸乙酯结晶,熔点110~111℃。略有吸湿性。溶于水。
用途
周围血管扩张药。
生产方法
糠醛和丙二酸二乙酯溶于甲苯,搅拌下加入适量催化剂,回流脱水。反应液水洗,浓缩减压蒸馏,得化合物(I),收率90%~95%(以丙二酸二乙酯计)。
化合物(I)和催化剂(Pd/C或Raney Ni),在80~100℃和1.5~4.5MPa氢压下氢化。降温,滤除催化剂,浓缩,得化合物(Ⅱ),产率90%~95%。
化合物(Ⅱ)和二乙氨基乙醇溶于甲苯,加入适量催化剂(如氢化钠、乙醇钠、钠或锂等碱金属化合物),反应一定时间后,减压蒸出甲苯和过量的二乙氨基乙醇,用碱水洗,得化合物(Ⅲ),收率85%~90%。
化合物(Ⅲ)溶于二甲基甲酰胺,剧烈搅拌下缓慢加入氢化钠,再30℃下滴加氯甲苯萘,55~60℃保温。除去二甲基甲酰胺,碱水洗,得化合物(Ⅳ),产率75%。
化合物(Ⅳ)溶于二甲基甲酰胺,加入水回流。蒸出二甲基甲酰胺,收集198~200℃/93.3Pa的馏分,得萘呋胺,产率约85%,纯度90%。往萘呋胺的丙酮溶液中,滴加草酸水合物的丙酮溶液,加热至沸,置于冰箱。滤集沉淀,干燥,用乙醇-乙酸乙酯重结晶,得草酸萘呋胺,熔点109~110℃。
