Middotolin (MS) is a structurally modified active derivative of staurosporine, an indole carbazole alkaloid isolated from Marine Streptomyces astrosporin in 1977, commonly known as 4 '-n-benzamido-Staurosporine in Chinese. The United States Food and Drug Administration (FDA) on February 19, 2016 granted the status of a breakthrough therapy for the treatment of adult AML patients newly diagnosed with FLT3 gene mutations when it is used in combination with other chemotherapeutic (chemotherapy) drugs, and granted fast-track review treatment. It was approved by the US FDA on April 28, 2017 under the product name Rydapt. It is also approved for the treatment of aggressive systemic mastocytosis (ASM), systemic mastocytosis with hematologic tumors (SM-AHN), and mast cell leukemia (MCL) in adults.
