Ethyl acetate is a carboxylic acid ester, a bio-friendly organic solvent with a wide range of industrial applications. It is a moderately polar compound containing a non-polar ethyl group, a polar carbonyl (C=O) group and a polar oxygen atom. The polarity of the carbonyl and oxygen groups gives ethyl acetate a degree of polarity that makes it a useful solvent for a variety of polar and non-polar compounds. It is commonly used as an extraction solvent in analytical chemistry for the separation and purification of analytes from complex matrices such as biological fluids, environmental samples and foods. Its low boiling point facilitates evaporation of solvents and recovery of analytes.

Ethyl acetate as a solvent in biosynthesis

Ethyl acetate is a commonly used solvent in biosynthesis, and is particularly suitable for the extraction and purification of small molecules such as natural products and metabolites from biological samples. Ethyl acetate is also the solvent of choice for certain enzyme-catalysed reactions, as it does not interfere with the reaction or affect enzyme stability. In addition, it is used for protein sequencing, peptide and oligonucleotide synthesis.

Ethyl acetate as mobile phase in chromatography

Ethyl acetate is a commonly used solvent in a variety of chromatographic techniques, including thin layer chromatography (TLC), high performance liquid chromatography (HPLC) and gas chromatography (GC). In TLC, it is commonly used as a component of the mobile phase to separate and characterise different components of a mixture. In HPLC and GC, ethyl acetate is used as a mobile phase solvent because it can efficiently solubilise a wide range of organic compounds and can be easily separated from the target analyte. Mixtures of ethyl acetate with methanol/ethanol are also used as mobile phases in HPLC. In addition, in TLC, for liquid-liquid extraction, ethyl acetate in hexane is used as the mobile phase.

Ethyl acetate is used in the pharmaceutical industry as a standard for various applications such as analytical testing and quality control, as well as for calibrating analytical instruments or validating analytical methods.

Labelled ethyl acetate (ethyl acetate-1-13C, ethyl acetate-2-13c) can be used to follow the metabolism of drugs in vivo or to study the mechanism of enzyme-catalysed reactions.
