Inhibitor of proteases and esterases
Phenylmethylsulfonyl fluoride is a potent inhibitor of proteases and esterases. It has been shown to have a complex activity in relation to organophosphorus delayed neuropathy (OPIDN), depending on the timing of its dosing relative to an inducer of delayed neuropathy. PMSF can either protect against OPIDN or potentiate/promote it. Studies have shown that PMSF acts by binding to various enzymatic entities, including Eα, Eβ, and Eγ, which have different sensitivities to PMSF and other inhibitors. Interestingly, preincubation with PMSF can alter the sensitivity of these enzymatic entities, with some becoming less sensitive to other inhibitors like mipafox and paraoxon. The exact nature of these interactions is not fully understood, but it is believed that PMSF may bind covalently to sites other than the substrate catalytic center. Inhibition of acetylcholinesterase

Monomeric AChE, on the other hand, was unaffected in all tissues examined. Furthermore, when Phenylmethylsulfonyl fluoride was added to a muscle homogenate in vitro, all forms of AChE were equally inhibited. In purified forms, both monomer and tetramer were equally susceptible to PMSF inhibition.
