11 Preparation of 1-methylbenzimidazole
Benzimidazole (100 mg, 0.85 mmol),DMF (4 mL),CH3ONa (137 mg, 2.5 mmol, 3 eq.) was added to the reaction flask in turn.After stirring at room temperature for 5 minutes, after NaH is uniformly dispersed,Slowly inject methyl trifluoroacetate (0.34 mL, 3.4 mmol, 4 eq.) into the reaction flask.After reacting for 10 hours, the TLC plate, the starting point disappeared, that is, the reaction was completed, and the stirring was stopped.Extract twice with ethyl acetate and distilled water.Wash with saturated NaCl solution and combine the organic phases.The organic phase was dried over anhydrous magnesium sulfate, and magnesium sulfate was removed by filtration.The brown liquid 1-methylbenzimidazole (81 mg, yield 72%) was purified by column chromatography.
