69.1 Step 1: 2-Amino-6-methoxynicotinic acid
To a solution of 2-amino-6-chloronicotinic acid (900 mg, 5.4 mmol) in 25 mL methanol was added sodium methoxide (2.81 g, 52.2 mmol). The resulting mixture was stirred at reflux for two weeks. After cooled to room temperature, the solvent was removed in vacuo and the residue was purified by C18 reverse phase column [Mobile Phase A: Water, Mobile Phase B: Acetonitrile; Gradient: 0% B to 60% B in 25 min] to give 156 mg of the title compound as a white solid. MS (ESIpos): m/z = 169 (M+H)+; LC-MS [Method 2]: R, = 0.66 min.
