Steps:

2.2.1. Synthesis of 2-amino-N-(4-methoxybenzyl)benzamide ( 3 )

Ethyl acetate (300 mL) and isatoic anhydride (50 g, 0.31 mol) were added to a 500 mL single-neck flask. Subsequently (4- methoxyphenyl)methanamine (40.04 mL, 0.31 mol) was slowly added under stirring at room temperature. After the addition was completed, temperature of the reaction solution was heated to 40 °C, and the reactional time was monitored by TLC using mixture of petroleum ether and ethyl acetate (1:1) and the plates were re- vealed with sublimated iodine supported on silica gel. After the re- action was stirred for 3 h, the mixture was cooled to room temper- ature. The reaction precipitated white solid and was filtered. The filter cake was slurried twice with water and methanol in turn. Af- ter the filter cake was dried to obtain the compound 3 (77.79 g, yield 99.1%) as white solid.
