Reaction Conditions:
Stage #1: 2-bromo-3-fluoroanisolewith n-butyllithium in diethyl ether at -78;
Stage #2: with sulfur dioxide in diethyl ether;
Stage #3: with N-chloro-succinimide in diethyl ether at 20 - 30;
Steps:
2.1 2-Fluoro-6-methoxybenzenesulfonyl chloride 2b
At -78°C, 2-bromo-3-fluoroanisole 2a (1.08g, 5.26mmol, Shaoyuan) was dissolved in anhydrous ether (20mL), and n-butyllithium (2.5M, 2.2mL, Bailingwei), reacted for 2 hours. Sulfur dioxide gas was passed through the reaction liquid for 30 minutes, N-chlorosuccinimide (777 mg, 5.81 mmol, Adamas) was added, and the reaction was carried out at room temperature for 1 hour. The reaction system was filtered, the filter cake was washed with dry ether, the filtrate was concentrated under reduced pressure, and the resulting residue was purified by silica gel column chromatography with eluent system B to obtain the title product 2b (400.0 mg, yield: 33.8%).
