42.1 Step-1: Synthesis of 3-(2-hydroxyethoxy)propanenitrile (13):
To a stirred mixture of ethane- 1,2-diol 12 (23.5 g, 379 mmol) and aq NaOH (30 mg), acrylonitrile (5.0 g, 94.0 mmol) was added and the reaction mixture was stirred at 45°C for 5h. The progress of the reaction was monitored by TLC. After completion of the reaction, the reaction mixture was concentrated under reduced pressure. The residue was neutralized with 6N HC1 solution and evaporated to dryness. The residue was purified by column (0825) chromatography using 90% EtOAc in hexane to afford 13. Yield: 8.6 g, 79.3%; 1H NMR (400 MHz, DMSO-i) δ 4.68 - 4.58 (m, 1H), 3.60-3.62 (m, 3H), 3.55 - 3.42 (m, 3H), 2.74- 2.76 (m, 2H).
