Steps:

Synthesis of 3-(4-methoxybenzyl)-2-thio-2,3-dihydroquinazolin-4(1H)-one

In a 1000mL single mouth flask, anhydrous ethanol (300 mL) and compound 2(77.79 g, 0.30 mol) were added, besides, potassium hydroxide (37.46 g, 0.67 mol) and carbondisulfide (181.96 mL, 3.04 mol) were added with stirring at room temperature, then the reaction solution was warmed up to 45 C. The reaction was monitored by TLC spot plate. After 10 h, the reaction mixture was cooled to room temperature and a solid was precipitated, then, it was filtered, the filter cake was slurried once with water and methy1 tert-buty1 ether, and dried to obtain a white solid of 82.31 g with a yield of 90.9%. 1H NMR (400 MHz, CDCl3) d (ppm) 10.13 (s, 1H), 8.13 (d, J7.9 Hz, 1H),2 L. YAO ET AL.7.64 (t, J7.6 Hz, 1H), 7.56 (d, J8.5 Hz, 2H), 7.31 (t, J7.6 Hz, 1H), 7.08 (d,J8.1 Hz, 1H), 6.83 (d, J8.5 Hz, 2H), and 5.73 (s, 2H), 3.77 (s, 3H).
