Dissolve benzo[4,5]thiophene[3,2-d]pyrimidin-4(3H)-one (4.38g, 21.6mmol) in 25mL of phosphorus oxychloride,Add two drops of N,N-dimethylformamide as a catalyst, heat to 110°C and reflux until the reaction is complete, about 5 hours (TLC monitoring).Cool the system to room temperature, slowly pour it into ice water, and neutralize it with sodium hydroxide to a pH of 8-9,Then the aqueous phase was extracted 3 times with ethyl acetate, the organic phases were combined, dried, and rotary evaporated.Separation by silica gel column chromatography (eluent: ethyl acetate: petroleum ether = 1:100, v/v),The white solid product 4-chloro-3,4-dihydrobenzo[4,5]thiophene[3,2-d]pyrimidine (4.75g, 99%) was obtained.
