Methyl 2-(4-methoxy-3-nitrophenyl)acetate (8.0 g, 35.6 mmol) was dissolved in tetrahydrofuran (80 mL) and water (20 mL). Lithium hydroxide monohydrate (4.5 g, 106.7 mmol) was slowly added to the reaction solution, and the reaction was stirred at room temperature for 1 hour. Tetrahydrofuran was removed under reduced pressure and the pH was adjusted to 3 with hydrochloric acid (3N). The reaction mixture was stirred for 15 minutes and then filtered. The filter cake was washed with 50 mL of water and dried to give 2-(4-methoxy-3-nitrophenypacetic acid. (6.0 g, 28.4 mmol, yield: 80%). 1H NMR (400 MHz, DMSO-d6) δ 12.46 (m, 1H), 7.79 (d, J=2.4 Hz, 1H), 7.56 (dd, J=2.4 and 8.8 Hz, 1H), 7.32 (d), J=8.8 Hz, 1H), 3.91(s, 3H), 3.64(s, 2H).
