A 100 mL three-necked reaction flask was added with 30 mL of phosphorus pentachloride, and 4-methoxy-2-picolinic acid (2b) (15.3 g, 100 mmol) was added in batches under stirring conditions, and the addition was completed in 10 minutes. After the addition, the reaction mixture was reacted in an oil bath at 100°C for 4 hours, and the phosphorus pentachloride was removed by distillation under reduced pressure, and 10 mL of toluene was added to the residue to dissolve. In addition, add 100mL to a 250mL three-necked reaction flask ammonia water (25-28% content), Cool in an ice-water bath, drop the above acid chloride solution at constant pressure, After dripping, react at room temperature for 1 hour. After the reaction, it was filtered, the solid was washed with 3×20 mL of water, and dried to obtain the solid product 4-methoxy-pyridine-2-carboxamide (3b) (14.2 g, yield 93%). The target product was confirmed by nuclear magnetic resonance spectroscopy.
