Steps:

3 6-methoxyquinoline-2-carboxylic acid (Compound 4)

A solution of 6-methoxyquinoline-2-carboaldehyde (Compound 3) (0.8 g, 4.27 mmol) dissolved in tert-butanol (20 mL) was carefully added dropwise to a solution of sodium dihydrogen phosphate (NaH₂PO₄; 4.10 g, 34.2 mmol) and sodium chlorite (NaClO₂, 3.87 g, 42.7 mmol) in water (10 mL), followed by allowing to react at room temperature for 2 hours. The reaction mixture was adjusted to a pH of 4 and extracted with ethyl acetate (3×20 mL), the organic layer was dried over sodium sulfate (Na₂SO₄), and the desiccant was removed by filtration. Thereafter, the filtrate was concentrated under reduced pressure, and the obtained product, Compound 4 (865 mg, 100%) was used in the following Reference Example 4 reaction without purification.
