2.10 Synthesis of Tert-butyl (4-(N'-hydroxycarbamimidoyl) phenyl] carbamate
To a suspension of tert-butyl(4-cyanophenyl) carbamate (1.0 equiv.) and hydroxylamine hydrochloride (1.5 equiv.) in EtOH (25 mL), NaHCO3 (1.5 equiv) was added. The mixture was stirred under reflux for 6 h. and eventually diluted with cold water (50 mL). the aqueous phase was extracted with ethyl acetate (3 x 50 mL). the combined organic phases were dried over Na2SO4, filtrated and concentrated. The residue was then purified on silica gel column chromatography eluting with 1-10 % methanol in dichloromethane to afford the desired product as white solid (1.12 g, 4.46 mmol, 97 %). 1H NMR (400 MHz. DMSO-d6): δ 9.48 (s, 1H,), 9.45 (s, 1H), 7.55 (d, J = 8.1 Hz, 2H), 7.43 (d, J = 8.1 Hz, 2H), 5.71 (s, 2H), 1.47 (s, 9H).
