Reaction Conditions:

with 1,1'-bis-(diphenylphosphino)ferrocene;palladium diacetate;triethylamine in N,N-dimethyl-formamide at 100; for 12 h;

Steps:

49.1 Step 1. Synthesis of methyl 1H-pyrazole [3,4-b] pyrimidine-5-carboxylic acid ester
5-bromo-1H-pyrazole [3,4-b] pyrimidine (5.0 g, 25 mmol), Pd (OAc) 2 (0.28 g, 1.25 mmol), dppf (1.4 g, 2.5 mmol), methanol (5.0 mL, 125 mmol) and Et3N (7.0 mL, 50 mmol) were added to a 100 mL round bottom flask, dissolved in DMF (15.0 mL), and replaced with CO three times. The reaction was carried out at 100 ° C for 12 hours. After the reaction was completed, it was cooled to room temperature, filtered, spin-dried, and separated by silica gel column chromatography. The obtained yellow solid was the target product (2.0 g, yield: 46%).
