Warfarin, 3-(α-acetonylbenzyl)-4-hydroxycoumarin (24.1.10), is synthesized via Michael reaction by attaching 4-hydroxycoumarin (24.1.7) to benzalacetone in the presence of pyridine.

Reaction Conditions:

in dimethyl sulfoxide at 20; for 16 h;

Steps:

4-hydroxycoumarin (compound 1, 1.62 g, 10 mmol),Benzylmethanol (Compound 2, 1.46 mg, 10 mmol)A solution of DL-valine (580 mg, 5 mmol) in DMSO (20 mL) was stirred at 20 ° C for 16 h.120 mL of water was added to the reaction mixture.The reaction mixture was extracted three times with ethyl acetate (100 mL).The combined organic phases were washed with 100 mL of water and brine (100 mL)The solvent was recovered on a rotary evaporator to give a white residue (3.3 g).The residue was separated by silica gel column chromatography ( petroleum ether: ethyl acetate = 4:1, v/v)A white powder (2.60 g, 84%) was obtained as Compound 3.
