异硫氰酸荧光素酯是黄色粉末，有吸湿性。其能与各种抗体蛋白结合，结合后的抗体不丧失与一定抗原结合的特异性，并在碱性溶液中仍有强烈绿色荧光，加酸后析出沉淀，荧光消失，微溶于丙酮、乙醚和石油醚。
