Preparation Method
The preparation method for N-(2-hydroxyethyl)-N-methyl-4-toluidine involves several steps and reagents to achieve a satisfactory yield of 70%. First, in a controlled environment such as a glove box, the reactant (N,4,4,5,5-Pentamethyl-N-(4-methylphenyl)-1,3,2-dioxaborolan-2-amine) is combined with dibromomethane and tetrahydrofuran (THF) in a Schlenk tube. This mixture is then cooled to -78°C. Next, butyllithium (nBuLi) is added dropwise under a nitrogen atmosphere, followed by stirring at -78°C for 30 minutes. Zinc chloride (ZnCl2) is then added, and the mixture is allowed to warm to room temperature and stirred for an additional hour. After cooling the mixture to 0°C, a premixture of hydrogen peroxide (H2O2) and sodium hydroxide (NaOH) is added, followed by stirring at room temperature for another hour. The mixture is then diluted with water and extracted with dichloromethane (DCM) before being dried over sodium sulfate. The crude product is then purified using silica gel flash column chromatography to obtain the desired product, N-(2-hydroxyethyl)-N-methyl-4-toluidine. This method involves alkylation/silylation of amines and phosphines, as well as coupling reactions such as alkyl/silyl halides coupling, Wurtz reaction, or Negishi coupling to achieve the desired chemical transformation. 3
