Long chain adipoxylic acid 18-(tert-butyl-18 oxyoctadecanoic acid) is often used in the synthesis of drugs, specialty materials, spices and a series of high value-added specialty chemicals. The synthesis of tert-butyl 17-carboxyheptadecanoate is a four step process which starts with reaction of octadecanedioic acid, concentrated sulfuric acid and methanol. The resulting product dimethyl octadecanedioate is reacted with barium hydroxide octahydrate and methanol, to produce Octadecane Diacid Alkane, followed with addition of Dichloromethane, concentrated sulfuric acid, finally Methanol, producing tert-butyl 17-carboxyheptadecanoate.

Steps:

1.B B. 18-(tertbutoxy)-18-oxooctadecanoic acid

A suspension of octadecanedioic acid (40 g, 127 mmol) in toluene (500 ml) was heated at 95 °C under nitrogen. To the resulting solution, was added N,N-dimethylformamide di- tert-butylacetal (98 g, 434 mmol), dropwise over 3-4 hr. The reaction was stirred overnight at the same temperature, concentrated to dryness in vacuo and placed under high vacuum overnight. The resulting solid was suspended in CH2Cl2 (200 ml) with heat and sonication, and filtered at RT, washing with CH2Cl2. The filtrate (2) was concentrated to give the product as a solid (45 g, 86 %) which was used without further purification.
